This class at ASU will cover practical methods in magnetic resonance. There will be hands-on NMR and MRI activities and students finishing the class will be able to do basic NMR and MRI experiments without supervision on both Varian and Bruker instruments.
NMR of Gramicidin-S (CHM 464) - “Biophysical Chemistry”
This 2-week laboratory exercise was developed for Biophysical Chemistry Lab by Prof. Jeff Yarger and Dr. Ron Nieman. Students should be able to determine the secondary structure of a small decapeptide, Gramicidin-S, purely from NMR experiments. The first week students run 1D 1H and 2D 1H-1H COSY and NOESY experiments on each individual amino acid and on the decapeptide. The second week students analyze data and learn to interpret 1D and 2D NMR spectra. The objective is to determine the primary and secondary structure of Gramicidin-S from the collected NMR data.

NMR Titration of Functional Groups in Proteins (Used in Analytical Chemistry Laboratory)

The paper describes how NMR can be used to simultaneously observe the protonation state of different ionizing functional groups in peptides and proteins. You can download the NMR spectra used in this study by clicking on the links below. The spectra were obtained at 400 MHz using a Varian UnityPlus spectrometer equipped with a 5mm pulsed-field gradient indirect-detection probe. Spectra for processing on unix, PCs or Macs have also been saved as compressed .zip files. Download the spectra and unzip them using something like winzip.exe, available from many download sites (e.g., www.tucows.com). You can use winzip.exe to download and extract the unix tar files on a pc. The standard compression utility for Macintosh, Stuffit Expander, will also handle both the compressed .zip.
